Coomassie Brilliant Blue (CBB) staining is one of the most widely adopted techniques for protein detection in polyacrylamide gels, particularly SDS-PAGE. Known for its simplicity, efficiency, and affordability, CBB staining remains a staple method in molecular biology and biochemistry laboratories.
The dye interacts primarily with basic amino acids such as arginine, lysine, and histidine, forming stable dye–protein complexes that appear as distinct blue protein bands against a clear gel background. The method allows reliable detection of proteins at levels as low as 30–100 ng, depending on the formulation.
